Di Stefano was born in Argentina in 1926 and began his football career with River Plate in 1945. He later played for several other Argentinian clubs before moving to Colombia to play for Millonarios in 1949.

He quickly became a star for the club, helping them win five consecutive European Cups from 1956 to 1960.
